About Semax
Semax is a seven residue peptide made by taking a four residue stretch of adrenocorticotropic hormone and appending the same proline, glycine, proline tail used in Selank. The parent fragment was chosen because published work indicated it carried behavioural activity without the hormonal activity of the full hormone. Like Selank, the compound comes out of Russian research institutes and most of its literature originates there.
The two compounds are worth reading side by side. They share a design principle and a stabilizing tail but start from entirely different parent molecules, one from an antibody fragment and one from a pituitary hormone. Whatever the behavioural literature shows, that shared approach is the clearest thing about them.

What it actually is
Adrenocorticotropic hormone is a 39 residue hormone whose main job is signalling to the adrenal glands. Researchers observed that a short internal stretch of it appeared to affect behaviour in animals without triggering the adrenal response, which suggested the two activities were separable. Semax is that stretch with a proline rich tail added so it survives long enough to be studied.

What the published work shows so far

Molecular Design and Stability
The sequence reads as two parts. Residues one to four, methionine, glutamate, histidine and phenylalanine, are the fragment taken from the hormone. Residues five to seven are the proline, glycine, proline tail added for resistance to enzymatic trimming. The methionine at position one is the most oxidation prone site on the molecule and is the reason dry, sealed storage matters more here than for peptides without sulphur containing residues.
The stabilizing tail does its work in solution against peptidases, but it does nothing about oxidation of the N terminal methionine, which is the practical concern for storage. Keep the vial sealed and dry until use, reconstitute the working volume, and do not store the material in solution.

What the Literature Covers

Expression changes are a measurement, not an outcome, and the distinction is routinely blurred in promotional writing about this compound.
3. Ischaemia models
A substantial part of the literature uses animal models of restricted cerebral blood flow. This is also the area behind the compound’s clinical registration in its country of origin. Model findings and a national registration are two different kinds of claim and neither is a general evidence base.
4. The same stabilizing tail as Selank
Both compounds end in proline, glycine, proline for the same reason. Reading them together makes the design strategy obvious and shows that the tail is a chemistry decision rather than anything to do with the activity of either parent fragment.
5. Where the evidence stops
