About TB-500
TB-500 is a short synthetic peptide built from the section of a larger protein called thymosin beta-4 that binds actin. Thymosin beta-4 itself is 43 residues long and occurs naturally in most human cells, where it is one of the proteins that governs how the internal scaffolding of a cell is assembled and taken apart. TB-500 reproduces only the seven residue stretch responsible for that binding, with an acetyl group on one end.
One point deserves stating plainly, because vendors routinely blur it. TB-500 and thymosin beta-4 are not the same molecule. Much of the published research described below was performed on the full 43 residue protein, not on the fragment. Where a study used the full protein, its findings belong to the full protein. TB-500 is the small piece of thymosin beta-4 that does the actual binding, synthesized on its own. It is far cheaper to make than the full protein and easier to handle, which is why it appears in laboratories at all.

Why researchers keep returning to it
For a laboratory studying cytoskeletal dynamics, a short peptide that binds actin is a useful and inexpensive probe. Molecular Design and Stability
The design question behind this peptide is whether a binding function spread across a large protein can be reduced to the handful of residues that actually perform it. Thymosin beta-4 binds actin through a short central motif, and TB-500 is that motif isolated and capped with an acetyl group at the N terminus. Acetylation removes a free positive charge and is a standard way to slow down the enzymes that trim peptides from that end.
At seven residues the molecule is small, water soluble and stable as a freeze-dried solid. In solution it behaves like any short peptide and should be treated as short-lived. Aliquot to the working volume rather than keeping a reconstituted vial.

The largest and best funded body of work sits in cardiac research, where thymosin beta-4 has been studied in rodent models of heart injury and in the biology of the epicardium. This work overwhelmingly used the full protein.
3. Corneal and dermal repair
A second cluster examines surface tissue, particularly the cornea, where the protein reached clinical investigation in its own right. Findings from those programmes belong to the full protein and to the specific formulation tested, not to a research fragment bought as a powder.
4. Vascular effects
Several groups report effects on endothelial cell behaviour and vessel formation in culture and in animals. The mechanisms proposed generally run through cytoskeletal reorganization, which is consistent with the actin binding role.
5. The fragment versus the protein
